Brucella sp. Real-time PCRProcedure: Real-time polymerase chain reaction (qPCR) analysis can be performed on DNA extracted from tissues, blood or fluids. This assay uses primers, probe and adapted protocols that target the gene for a 31 kDa outer membrane protein bcsp31, which is specific to genus Brucella (Probert et. al, 2004. Journal of Clinical Microbiology 42(3):1290.) The assay also includes two internal controls to assess DNA quality and presence of PCR inhibitors. Samples accepted: tissues (spleen, pulmonary and inguinal lymph nodes, tonsil, lung, liver, kidney, brain, gonad, uterus or epididymis), whole blood or buffy coat, other body fluids Specimen: small tissue samples (0.5 cm3) are required, unless simultaneous culture is desired (>2.5 cm3); >200 µl EDTA-preserved (not heparin) whole blood/buffy coat or other body fluids Sample storage: Short-term storage at -20°C is permissible, however if long term storage is necessary, tissues should be frozen at -70°C. Blood and fluids are ideally submitted fresh/cooled, but may be frozen. Sample shipment: overnight on ice packs (for fresh/blood samples) or frozen, preferably on dry ice Test days: when sufficient numbers of samples are received Minimum Test time: 5 days
