Available Tests:

Brucella sp. Real-time PCR

Procedure: Real-time polymerase chain reaction (qPCR) analysis can be performed on DNA extracted from tissues, blood or fluids. This assay uses primers, probe and adapted protocols that target the gene for a 31 kDa outer membrane protein bcsp31, which is specific to genus Brucella (Probert et. al, 2004. Journal of Clinical Microbiology 42(3):1290.) The assay also includes two internal controls to assess DNA quality and presence of PCR inhibitors.

Samples accepted: tissues (spleen, pulmonary and inguinal lymph nodes, tonsil, lung, liver, kidney, brain, gonad, uterus or epididymis), whole blood or buffy coat, other body fluids

Specimen: small tissue samples (0.5 cm3) are required, unless simultaneous culture is desired (>2.5 cm3); >200 µl EDTA-preserved (not heparin) whole blood/buffy coat or other body fluids

Sample storage: Short-term storage at -20°C is permissible, however if long term storage is necessary, tissues should be frozen at -70°C. Blood and fluids are ideally submitted fresh/cooled, but may be frozen.

Sample shipment: overnight on ice packs (for fresh/blood samples) or frozen, preferably on dry ice

Test days: when sufficient numbers of samples are received

Minimum Test time: 5 days

DISCLAIMER
This is a research-based test that is currently under development and validation. Therefore, the results of this assay should be interpreted with caution. Performance characteristics of this assay are yet to be determined under all conditions and clinical circumstances. Results should be interpreted in the context of each case, given the totality of clinical and diagnostic test findings. Negative results do not guarantee the absence of infection. Additional information is needed about the nature of marine Brucella before further interpretations can be made. It is up to the discretion of each submitting facility to decide how to use the results of these tests and to determine if further testing on specific animals is warranted.
